Natural product-based PD-1 pathway blockade for Human cancer therapy: Avacado (Persea Americana var. Introduction:What they say

A recent study from Division of Infectious Diseases, School of Medicine, Johns Hopkins University, Baltimore, MD 21218, United States shows that “TGF-β1-Mediated Smad3 Enhances PD-1 Expression on Antigen-Specific T Cells in Cancer.” This study was published in the 28 September 2016 issue of Cancer Discovery (one of the best journals in Cancer Science with an impact factor of 19+) by Prof Andrea L. Cox, Park BV and others.

On the foundation of this interesting finding, Dr L Boominathan PhD, Director-cum-chief Scientist of GBMD, reports that: Natural product-based PD-1 pathway blockade for Human cancer therapy: Avacado (Persea Americana var. Hass ) extract inhibits the expression of co-inhibitory receptor PD-1 on Antigen- Specific T-cells, increases the production of cytokines by tumor-infiltrating lymphocytes (TIL), and draining lymph nodes and augments anti-tumor activity via down-regulation of its target gene

What is known?

It has recently been shown that blocking PD-1 with antibodies one could make tumors shrink. This work, relating to Cancer immunotherapy, has been chosen as Science’s breakthrough of the year recently.

What we say: Research Findings

This study suggests a natural product-based Human cancer therapy.

Figure 1  Avacado fruit extract inhibits pulmonary tuberculosis via down-regulation of (Toll-like receptor-TLR) TLR-2 and Chemokine CXCL5 and up-regulation of phosphatase MKP-1

Figure 1  Avacado fruit extract   suppresses the expression of PD-1 and augments anti-tumor immunity

The study presented here suggests that Avacado (Persea Americana var. Hass) extract, by decreasing the expression of its target gene, it may suppress the expression of PD-1. Thereby, it may (1) increase the production of cytokines by tumor-infiltrating lymphocytes (TIL), and draining lymph nodes; (2) augment anti-tumor activity of the immune system; & (3) inhibit metastatic cancer progression.

Therapeutic opportunity

Thus, pharmacological formulations encompassing “Avacado extract or a pharmacologically active compound isolated from it” may be used to inhibit the progression of tumors.
