Natural product-based PD-1 pathway blockade for Human cancer therapy: Epigallocatechin-3-gallate, one of the major constituents of green tea, inhibits the expression of co-inhibitory receptor PD-1 on Antigen- Specific T-cells, increases the production of cytokines by tumor-infiltrating lymphocytes (TIL), and draining lymph nodes and augments anti-tumor activity via down-regulation of its target gene, 3/November/2016, 8.38 am

Introduction:What they say:

A recent study from Division of Infectious Diseases, School of Medicine, Johns Hopkins University, Baltimore, MD 21218, United States shows that “TGF-β1-Mediated Smad3 Enhances PD-1 Expression on Antigen-Specific T Cells in Cancer.” This study was published in the 28 September 2016 issue of Cancer Discovery (one of the best journals in Cancer Science with an impact factor of 19+) by Prof Andrea L. Cox, Park BV and others.

What we say:

On the foundation of this interesting finding, Dr L Boominathan PhD, Director-cum-chief Scientist of GBMD, reports that: Natural product-based PD-1 pathway blockade for Human cancer therapy: Epigallocatechin-3-gallate, one of the major constituents of green tea, inhibits the expression of co-inhibitory receptor PD-1 on Antigen- Specific T-cells, increases the production of cytokines by tumor-infiltrating lymphocytes (TIL), and draining lymph nodes and augments anti-tumor activity via down-regulation of its target gene

What is known?

It has recently been shown that blocking PD-1 with antibodies one could make tumors shrink. This work, relating to Cancer immunotherapy, has been chosen as Science’s breakthrough of the year recently.

Research Findings

This study suggests a natural product-based Human cancer therapy.

Green tea extract slows down aging and increases median lifespan via up-regulation of BuBR1

Epigallocatechin-3-gallate

The study presented here suggests that Epigallocatechin-3-gallate, by decreasing the expression of its target gene, it may suppress the expression of PD-1. Thereby, it may (1) increase the production of cytokines by tumor-infiltrating lymphocytes (TIL), and draining lymph nodes; (2) augment anti-tumor activity of the immune system; & (3) inhibit metastatic cancer progression.

Therapeutic opportunity:

Thus, pharmacological formulations encompassing “Epigallocatechin-3-gallate or its analog” may be used to inhibit the progression of tumors.
