Natural product-derived Life-extension therapy: Celastrol, found in the root extracts of Tripterygium wilfordii and Celastrus regelii, inhibits the expression of Ribosomal protein S6 kinase 1 (p70S6 Kinase) and extends mammalian life span, 27/December/2016, 11. 45 pm

Molecular therapy for N-Myc-overexpressing tumors: BMP4 increases the levels of tumor suppressor proteins, TAp63, TAp63 and Fbw7 (SCF Fbw7 ubiquitin ligase), inhibits the expression of pro-survival proteins N-Myc, Cyclin-E and Mcl-1, and stalls the progression of N-Myc-overexpressing cancers via down regulation of its target gene, 27/December/2015, 11.08 pm

What they say?

A recent study from the Institute of Healthy Ageing, Centre for Diabetes and Endocrinology, Department of Medicine, University College London, London WC1E 6JJ, UK shows that “Ribosomal protein S6 kinase 1 signaling regulates mammalian life span.” This study was published in the 2 October 2009 issue of the Journal “Science” (I.F: 31.477 ) by Prof Dominic J. Withers, Dr. Selman C and others.

What we say

On the foundation of this interesting finding, Dr L Boominathan PhD, Director-cum-chief Scientist of GBMDreports, for the first time, that: Natural product-derived Life-extension therapy: Celastrol, found in the root extracts of Tripterygium wilfordii and Celastrus regelii, inhibits the expression of Ribosomal protein S6 kinase 1 (p70S6 Kinase) and extends mammalian life span

Figure 1. Celastrol functions as a longevity promoter by decreasing the expression of p70 s6 kInase

This study suggests an miRNA-based life extension therapy. Celastrol, by  decreasing  the expression of its target gene, it may suppress Ribosomal protein S6 kinase 1 (p70S6 Kinase) expression. Thereby, it may promote: (1) resistance to bone, immune, and motor dysfunction; (2) insulin sensitivity; and (3) longevity (figs. 1 and 2).

Thus, pharmacological formulations encompassing Celastrol or its analogues or Celastrol in combinationwith any of the known longevity-promoting compound(s) can be used to extend the lifespan of an individual (fig. 2).
