Significance: This study provides mechanistic insights into how Vitamin-D inhibits cancer development. 1,25-dihydroxyvitamin D, by down regulating the expression of its target gene, it may promote miRNA biogenesis. Thereby, it may inhibit cancer development. Given that sunlight boosts the production of Vitamin D, exposing one to the optimal level of sunlight may inhibit cancer development by increasing the expression of tumor suppressor genes. Together, this study suggests, for the first time, that (1) 1,25-dihydroxyvitamin D may augment the production of small RNAs; and (2) pharmacological formulations encompassing “1,25-dihydroxyvitamin D or its analogues“ may inhibit cancer development by turning on the expression of tumor suppressor miRNAs.
