Significance: This study suggests, for the first time, that MiRNA-1269, by decreasing the expression of its target gene, may: (1) decrease the expression of gluconeogenesis genes, such as PEPCK and G6Pase; (2) reduce hepatic glucose production; (3) improve muscle insulin sensitivity; (4) decrease white adipose tissue mass and adiposisty; and (5) mimick the positive effects of exercise. Thus, pharmacological formulations encompassing “MiRNA-1269 or its activators” may be used to inhibit insulin resistance, decrease adiposity, and treat T2DM.
