Significance: This study suggests, for the first time, that Agaricus blazei Murill, by decreasing the expression of its target gene, it may (1) promote the tumor suppressor function of TAp73; (2) increasee senescence, and; (3) inhibit the expression of cancer stem cell genes. Given that tumor suppressor p53 is mutated in more than 50% of different human cancers, this study suggests a therapeutic strategy as to how mutant p53 expressing human cancers can be cured; that is, by activating its un-mutated homologous proteins, such as p73, in cancer cells. Thus, pharmacological formulations encompassing “Agaricus blazei Murill or small molecule compounds isolated from Agaricus blazei Murill” may be used to inhibit the progression of human tumors, including p53-mutated human tumors.
