Significance:  It has been known for years that Vitamin-D3 promotes insulin sensitivity. However, the mechanistic basis of this is far from clear.  This study suggests, for the first time, that  1,25-dihydroxyvitamin D3, by increasing the expression of its target gene, it may decrease the expression of a number of stress-related proteins. Thereby, it may increase the insulin-secreting cell mass and function.  Given the mechanistic basis, clinicians may consider prescribing 1,25-dihydroxyvitamin D3 to treat DM.
