Introduction: What they say
A study from Faculty of Medicine, Friedrich-Schiller-University, Germany shows that “Epigenetic stress responses induce muscle stem-cell ageing by Hoxa9 developmental signals.” This research paper was published in the 30 November 2016 issue of the journal “Nature” [One of the best research journals in General Science with an I.F of 38+] by Prof.Lenhard Rudolph, Stefan Tümpel, Simon Schwörer and others.
What we say:
On the foundation of this interesting finding, Dr L Boominathan PhD, Director-cum-chief Scientist of GBMD, reports that: Natural product-derived regenerative therapy for improving muscle repair and strength in the elderly: AbM (Agaricus blazei Murill) decreases the expression of HOXA9, increases skeletal muscle regeneration and improves muscle stem cells function and repair via up regulation of its target genes
What is known?
Prof. Lenhard Rudolph’s research team has recently shown that deletion of HOXA9 in mice results in a) down regulation of inhibitors of muscle stem cells function, including Wnt, TGFß, JAK/STAT and senescence signalling; and b) improved muscle stem cells regeneration and function. Conversely, overexpression of HOXA9 has been shown to promote ageing-associated features in muscle stems cells.
What we said earlier?
I had shown earlier that Agaricus blazei Murill (AbM): (1) inhibits the expression of co-inhibitory receptor PD-1 on Antigen- Specific T-cells, increases the production of cytokines by tumor-infiltrating lymphocytes (TIL) and draining lymph nodes and augments anti-tumor activity; (2) promotes NK cell maturation and function; (3) suppresses the expression of Tripeptidyl Peptidase 1 (TPP1) and inhibits cancer progression; (4) inhibits epithelial-mesenchymal transition, glycolysis and cancer progression via down regulation of GLUT3 and PDK1; (5) inhibits metastatic adenocarcinoma via down regulation of Lin28; (6) inhibits the development of diabetic cardiomyopathy; and (7) extends mammalian life span by down regulating the expression of Insulin receptor substrate-2 and p70 S6 kinase.
From Research findings to Therapeutic opportunity:
This study suggests, for the first time, a natural product-based regenerative therapy for improving muscle strength and repair in the elderly. AbM, by increasing the expression of its target gene, it may decrease the levels of HOXA9 (fig.1).

Thereby, it may: (1) down regulate inhibitors of muscle stem cells function; and (2) improve of muscle stem cells regeneration and function. Thus, pharmacological formulations encompassing “AbM or an active compound isolated from it or AbM plus any of the known compounds that promote muscle regeneration” may be used to suppress muscle stem-cell ageing, age-associated physiological decline and improve health span.
