A study from the Center for Inflammation, Immunity & Infection, Institute for Biomedical Sciences, Georgia State University, Atlanta, GA, 30303, USA shows that: “Curcumin suppresses NTHi-induced CXCL5 expression via inhibition of positive IKKß pathway and up-regulation of negative MKP-1 pathway.” This study was published in the 19 August 2016 issue of the journal “Scientific reports” by Prof. Li JD, Konduru and others.
On the foundation of this interesting finding, Dr L Boominathan PhD, Director-cum-chief Scientist of GBMD, reports that: Natural product therapy for Otitis Media: α-Lipoic acid, found in broccoli, yams, tomatoes, Brussels sprouts etc., induces MKP-1 expression, induces MKP-1 expression, inhibits IKKß phosphorylation, suppresses NDHi-induced inflammatory mediator C-X-C motif chemokine ligand 5 (CXCL5) expression, controls overreactive inflammatory responses, inhibits the progression of Otitis media and ameliorates conductive hearing loss via up-regulation of its target gene
Significance:
Given that: (1) Ottis media is an infection/inflammation of middle ear, with debilitating consequences of conductive hearing loss; (2) molecular mechanisms involved in the development of middle ear inflammatory diseases is poorly understood; (3) drugs that are currently used to treat inflammatory disease of ear are ineffective; (4) Ottis media affects about 16% of people (about 740 million) each year and it is more common amoung children. (5) nearly 141 million people have hearing loss due to ottis media globally each year; (6) Nontypeable Haemophilus influenzae (NTHi) is the major bacterial pathogen that causes Ottis media; (7) NTHi that causes middle ear infections become resistant to conventional antibiotic therapy frequently; (8) prolonged use of immunosuppressant drugs is detrimental to the health of childrens; and (9) millions of dollars are being spent every year worldwide to treat the infections/inflammation of ear, there is an urgent need to find: (i) a way to control abnormal levels of inflammatory mediators found in ottis media patients; (ii) non-antibiotic/steroidic therapeutics; (iii) a side-effect-free natural product-based drug; and (iv) a way to effectively treat and cure Ottis media.
Prof. Li’s research team has recently shown that Curcumin, by suppressing the expression of CXCL5, it inhibits NTH-1-induced Ottis media.
This study suggests an yet another natural product-based compound for the treatment of Ottis media.

This study suggests, for the first time, that α-Lipoic acid, by increasing the expression of its target gene, it may: (1) increase the expression of MKP/DUSP1; (2) inhibit IKKß phosphorylation; (3) suppress NDHi-induced inflammatory mediator C-X-C motif chemokine ligand 5 (CXCL5) expression; and (4) control overreactive inflammatory responses. Thereby, it may: (1) inhibit the progression of Otitis media; and (2) ameliorate conductive hearing loss. Thus, pharmacological formulations encompassing “α-Lipoic acid or its analogues” may be used to treat Ottis media.
